Headliners include the Kaiser Chiefs, Scouting For Girls, Groove Armada, Republica, East 17, and Artful Dodger.
Kaiser Chiefs bassist Simon Rix said: "We're looking forward to it.
"We've got a few gigs leading up to that so we should be well in our stride by then – plus, we're playing on Sunday night; there's no excuses for not having a good time.
"We'll definitely have a good time."
The festival began as a community event and has grown into a major fixture on the UK summer festival circuit.
Friday’s opening night will feature tribute acts Fu Fighters and Coldplace, while Saturday will see Groove Armada’s DJ set, The Fratellis, and Heather Small.
Sunday’s line-up includes Kaiser Chiefs, Scouting For Girls, East 17, and Dodgy.
The full weekend will be packed with entertainment on four stages, spanning both the best of the charts and top tribute performances.
The event begins at 5pm on Friday, and noon on Saturday and Sunday.
It will close at 11pm on Friday and Saturday, and 8pm on Sunday.
Tickets and further details are available at the Poole Harbour Festival website.
